Two-dimensional gel electrophoresis (2D-PAGE)


Two-dimensional gel electrophoresis is combined with isoelectric focusing (IEF) and SDS-PAGE methods to enhance the resolution of the electrophoresis.
The separation of different proteins with two-dimensional gel electrophoresis is based on the isoelectric point and the molecular point of different proteins. With two different separating methods, we are able to identify and quantify thousands of proteins.

We use IEF to perform one-dimension gel electrophoresis and separate the proteins according to their PI. Then we use SDS-PAGE to perform the two-dimension gel electrophoresis and separate the proteins according to their molecular weight.
With this method, we can identify the protein composition in a complicated sample, or analyze the difference of protein composition with different treatments.

Staining method: 
1. Coomassie blue stain
2. Silver stain
